Essay

1-Trifluoromethyl-1-aryl/heteroaryl-disubstituted cyclopropanes and cyclobutanes. Development of efficient and scalable synthetic methods for these compounds, investigation of their physico-chemical and biological properties, and evaluation of their potential as structural analogs of the tert-butyl group in biologically active molecules. Organic synthesis; instrumental techniques for structural elucidation and purity confirmation of synthesized compounds (¹H, ¹³C, and ¹⁹F NMR spectroscopy, mass spectrometry (MS), high-resolution mass spectrometry (HRMS), elemental analysis, X-ray crystallography); biological activity studies. For the first time, a systematic investigation of the deoxofluorination reaction of cyclopropanecarboxylic and cyclobutanecarboxylic acids has been conducted, and a preparative method for multigram-scale synthesis of new compounds containing CF₃-cyclopropane and CF₃-cyclobutane fragments has been developed. The ability of alkali metal salts of carboxylic acids to also undergo deoxofluorination reactions has been demonstrated. Possible modification pathways of the synthesized compounds have been explored. It has been shown that 1-CF₃-1-disubstituted cyclopropanes and cyclobutanes are promising structural motifs in bioactive compounds as bioisosteres of the tert-butyl group.
